To improve the application of endophyte Bacillus velezensis BY6 from the xylem of poplar, the effect of BY6 on the growth of diseased Populus davidiana × Populus. alba var. pyramidalis Louche (Pdpap poplar) seedlings and the biological control effect on the pathogen Armillaria solidipes were tested using a plant split-root experiment. After applying BY6 to the roots of diseased Pdpap poplar seedlings, the results show that plant growth indicators (dry mass, fresh mass, and plant height) were significantly increased (p < 0.05), and genes related to auxin hormone signal transcription were activated. BY6 indicated a surprising control effect after the inoculation of diseased Pdpap poplar seedlings. Compared to the infected control group, the treated disease index of the diseased Pdpap poplar seedlings in the treatment group were reduced by 49.53% on the 20th day. The relative staining areas of diaminobenzidine (DAB) and Trypan blue decreased by 3.37 and 7.31 times, respectively. The physiological indicators (soluble sugar and protein) and oxidase indicators were significantly increased (p < 0.05). The expression levels of defense genes related to salicylic acid (SA) and jasmonic acid (JA) signaling pathways were significantly increased (p < 0.05). Amazingly, the results indicate that BY6 simultaneously activates induced systemic resistance (ISR) and systemic acquired resistance (SAR) in diseased Pdpap poplar seedlings and promotes growth. The results indicate that BY6 is a promising candidate for developing forest tree biofertilizers and biopesticides.
Phosphorus (P) is a critical nutrient for plants, and inorganic orthophosphate (Pi) deficiency results in declining crop quality and yields. Auxin plays a key regulatory effect on plant growth in response to Pi deficiency, yet the regulatory mechanisms remain poorly understood. In this study, we cloned and identified an auxin efflux transporter, NtPIN3, involved in the low phosphate response of tobacco (Nicotiana tabacum). NtPIN3 is located on the cytoplasmic membrane, and high expression levels of NtPIN3 were observed under phosphorus deficiency conditions. Moreover, NtPIN3 overexpression in tobacco could dramatically enhance low Pi tolerance by increasing the root length, which was consistent with the GUS activity and the auxin concentration in the root. Notably, Pi concentration and antioxidant capacity in overexpression transgenic tobacco were significantly increased compared with the wild type (WT) under low Pi stress conditions due to Pi uptake (NtPT1 and NtPT2) and antioxidant capacity (superoxide dismutase, peroxidase, and catalase). In contrast, compared with WT, the ntpin3 mutant significantly reduced root length, auxin concentration, Pi concentration, and antioxidant capacity under low Pi stress conditions, suggesting that NtPIN3 is a positive regulator of low phosphate in tobacco. In addition, the yeast-two-hybrid and luciferase complementation imaging assay confirmed that NtPIN3 interacted with NtPIN2. Altogether, the NtPIN3 involved in low Pi response and overexpression of NtPIN3 contributed to enhanced tolerance to low Pi stress, which proved an underlying gene for breeding low Pi-stress tolerant plants.

The fusion of aesthetic education with vocational training constitutes a necessary evolution dictated by both societal progression and the demands of contemporary times, with each element enhancing the other. This study proposes a pathway for the construction of aesthetic education courses tailored to the current educational objectives of vocational undergraduate institutions. It also introduces a teaching model designed to foster aesthetic sensibilities structured around the ADDIE instructional design framework. Moreover, this research employs the 2023 student cohort from HZ Vocational College as subjects for a comparative experimental study to empirically analyze the effectiveness of the aesthetic education teaching approach. An enhanced clustering algorithm, which integrates multi-point optimization, was applied to assess the development of students’ aesthetic spirit. Additionally, a regression analysis model was used to investigate the various factors influencing the cultivation of this aesthetic spirit, providing a comprehensive analysis of the impact of the proposed educational interventions. The results show that after the teaching, the mean values of student evaluation, self-evaluation, parent evaluation, and teacher evaluation in the five aspects of recognizing beauty, experiencing beauty, feeling beauty, appreciating beauty, and creating the beauty of the students in Class A of the sample are higher than those of Class B by 4.90, 4.08, 3.40 and 4.13. Their p-values are all statistically significant at 0.000<0.05. The ADDIE-based aesthetic education teaching model can effectively cultivate students’ aesthetic spirit and provide important value references for the reform and development of aesthetic education.
Objective FCM(flow cytometry) is compared with Western blot in investigating the phosphorylated of STAT3.Methods Promyelocytic leukemia cell line HL-60 are cultured and stimulate with human IL(hIL)-5 in different concentration and then are detected with Western Blot and FCM simultaneously.Results The results of Western blot and FCM are identical.Conclusion The two methods are all have their own advantages in investigating phosphorylated STAT3 and they are mutually complementary.
This paper introduces the design and realization of the Multi-function Tower Crane Black Box, completes the main hardware module design and type selection. With ZigBee, it constructing wireless sensor network to collect the communication among nodes and the communication between nodes and the gateway, the lower PC adopts the IAR to complete the chip CC2430 programming. The application development tool for the upper PC is C51RF - WSN - DA300, and it gives part of the source code. The system parameters acquisition is accurate, the operation is stable, and the actual application effect is good.
