A series of novel analogs of acyclovir, substituted with an alkyl (methyl, ethyl, n-butyl) or phenyl group at the positions 1', 4', and/or 5', has been obtained in a direct one-pot coupling reaction of guanosine and the respective 1,3-dioxolanes. The new acyclonucleosides were essentially inactive in antiviral (HSV, VV, VSV, HBV) evaluation in vitro.

Sodium 2-mercaptoethanesulfonate reacts with the metal ions Pd(II), Pt(II), Ag(I), Cd(II) and Zn(II) to yield complexes containing multiple anionic sulfonate sites. On the basis of spectroscopic and other analytical data the complexes were assigned the tentative molecular formulas: Pd6(SCH2CH2SO3Na)12, Ptn(SCH2CH2SO3Na)2n+2, Agn(SCH2CH2SO3Na)n, Na2Zn4(SCH2CH2SO3Na)10, and Na2Cd4(SCH2CH2SO3Na)10. The complexes displayed a variety of differences in activity towards DNA and RNA viruses. The platinum complex showed no measurable cytotoxicity and exhibited a spectrum of antiviral activity resembling that of dextran sulfate. It was active against HIV-1 and HIV-2, herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2), thymidine kinase-deficient HSV-1, human cytomegalovirus, vesicular stomatitis virus (VSV), influenza A virus, respiratory syncytial virus (RSV), Sindbis virus, Junin virus and Tacaribe virus. The palladium complex also showed no measurable cytotoxicity, but was completely inactive against most viruses, with one notable exception: both HIV-1 and HIV-2 were substantially inhibited by the palladium complex. The silver complex showed significantly less antiviral activity and greater cytotoxicity than the platinum complex but did show some selectivity against RSV. The zinc complex showed only modest activity against VSV, RSV, Junin virus, and Tacaribe virus, and like the silver compound was more cytotoxic than either the platinum or palladium complex. The cadmium complex was toxic to all of the cell lines used for in vitro evaluation of antiviral activity. Based on these results, the platinum and palladium compounds appear to be promising candidates for further studies, that is, as vaginal microbicides in the prevention of genital HIV and/or HSV transmission.

Dextran sulfate and pentosan polysulfate are two promising candidate anti-acquired immune deficiency syndrome (AIDS) drugs that have already been the subject of initial clinical trials in patients with AIDS. There is at present no reliable assay method to monitor blood drug levels in humans following the administration of either drug. We have now developed a sensitive bioassay system based on the inhibitory activity of the compounds against human immunodeficiency virus type 2 (HIV-2) in MT-4 cells. This method permits the detection in (rabbit) serum samples of dextran sulfate and pentosan polysulfate concentrations as low as 3.0 and 0.5 micrograms/ml, respectively. Pharmacokinetic studies with dextran sulfate and pentosan polysulfate in rabbits indicated that the half-life of these compounds after intravenous bolus injection is approximately 1 h 24 min.

This paper describes the influence of dynamic tension/compression loading on notched and unnotched nylon specimens fabricated by Injection Molding (IM) and Selective Laser Sintering (SLS). The main objective of this work is to analyze and describe the differences in material structure and fatigue properties of as-built nylon parts produced by IM and SLM from the same polyamide 12 powder. The differences in dimensional quality, density, surface roughness, crystal structure and crystallinity are systematically measured and linked to the mechanical fatigue properties. The fatigue properties of the unnotched SLS specimens are found to be equal to those of the unnotched IM specimens. The presence of pores in the sintered samples does not lead to rapid failure, and the microvoid coalescence failure mechanism is delayed. The notched specimens show more brittle failure and increased fatigue resistance which is caused by local notch-strengthening. The results enable improved understanding of the difference in material structure and fatigue behavior of selective laser sintered and injection molded polyamide.

A group of unnatural 1-(2-deoxy-beta-D-ribofuranosyl)-2,4-difluorobenzenes having a variety of C-5 two-carbon substituents [-C...C-X, X = I, Br; -C...CH; (E)-CH=CH-X, X = I, Br; -CH=CH2; -CH2CH3; -CH(N3) CH2Br], designed as nucleoside mimics, were synthesized for evaluation as anticancer and antiviral agents. The 5-substituted (E)-CH=CH-I and -CH2CH3 compounds exhibited negligible cytotoxicity in a MTT assay (CC50 = 10(-3) to 10(-4)M range), relative to thymidine (CC50 = 10(-3) to 10(-5)M range), against a variety of cancer cell lines. In contrast, the C-5 substituted -C...C-I and -CH(N3)CH2Br compounds were more cytotoxic (CC50 = 10(-5) to 10(-6)M range). The -C...C-I and -CH2CH3 compounds exhibited similar cytotoxicity against non-transfected (KBALB, 143B) and HSV-1 TK+ gene transfected (KBALB-STK, 143B-LTK) cancer cell lines expressing the herpes simplex virus type 1 (HSV-1) thymidine kinase gene (TK+). This observation indicates that expression of the viral TK enzyme did not provide a gene therapeutic effect. The parent group of 5-substituted compounds, that were evaluated using a wide variety of antiviral assay systems [HSV-1, HSV-2, varicella-zoster virus (VZV), vaccinia virus, vesicular stomatitis, cytomegalovirus (CMV), and human immunodeficiency (HIV-1, HIV-2) viruses], showed that this class of unnatural C-aryl nucleoside mimics are inactive and/or weakly active antiviral agents.

Large chromosomal DNA fragments containing different parts of the putative rubromycin polyketide synthase gene cluster were cloned and functionally expressed in S. coelicolor CH999. Expression of these clones yielded 5 approximately 10 metabolites that were not detected in S. collinus culture extracts. This paper focusses on one of the new metabolites, termed collinone, that was isolated in large quantities and purified for spectroscopic structure determination and biological screening assays. Collinone is a heavily oxidized angular hexacyclic compound containing an unusual 1,4,5,8(2H,3H)-anthracenetetrone moiety previously only reported to be present in antibiotics SF2446A1, A2, A3, B1 and B2 isolated from Streptomyces sp. SF2446. Structure analysis of collinone indicates a tridecaketide with a 26 carbon backbone. The basic benz[a]naphthacene ring system of collinone is angular, similar to the aglycones of the well-known angucycline and angucyclinone antibiotics. While collinone showed antibacterial activity against vancomycin-resistant enterococci, no antifungal or significant antiviral activities were detected. Collinone could be a good starting point to obtain new bioactive angucyclin(on)e-like compounds by further genetic engineering of its pathway.

Ever since composite materials where first introduced, they have been pushing the boundaries of high performance, lightweight designs in all branches of engineering. The demand for sustainable lightweight structures results in an augmented use of thermoplastic composites. Depending on the type of matrix and reinforcement, there are various manufacturing options for the fabrication of composite parts. Composite manufacturing processes are in essence additive processes. In order to reduce the labor-intensive manual operations, and the need for a flexible automated composite process, researchers are investigating the feasibility of implementing Additive Manufacturing (AM) techniques to aid the fabrication of composite parts. AM techniques are able to produce parts directly from CAD data sources. As opposed to classical subtractive fabrication methods, parts are created layer upon layer. The geometric freedom provided by the additive process unlocks a wide variety of designs, which would be impossible to create via subtractive methods. Furthermore, AM processes have no direct need of tooling. The flexibility of this manufacturing approach gives rise to the development of application-oriented parts. Given the flexibility of the additive process, these techniques can be used in the design and manufacturing of composite parts. There are several options for which AM can be implemented in the composite production process. This paper highlights the potential of AM in the design and manufacturing of composite parts, gives a review on the application of composite AM, and identifies the technological challenges associated with the direct production of thermoplastic AM composites.

This paper provides new insights into the fatigue properties of Selective Laser Molten components made from Ti6Al4V powder particles. The SLM-process parameters are optimized and high quality SLM-parts with a relative density of 99.7% are produced. Uniaxial fatigue experiments are then performed on notched and unnotched specimens and the endurance limits are determined using the staircase method and the theory of Dixon and Mood. The presented results indicate inferior fatigue strength in comparison with conventionally produced components from Ti6Al4V. Microstructural analysis shows that this is mainly due to the anisotropy in the microstructure and the weak grain boundaries between epitaxial grains. However, the failure mechanism is consistant leading to low statistical scatter in the fatigue data. Furthermore, the critical volume method and the critical distance theory have proven to be accurate and efficient design tools to account for the notch-effect in SLM-Ti6Al4V.

There is an urgent unmet medical need for novel human immunodeficiency virus type 1 (HIV-1) inhibitors that are effective against a variety of NNRTI-resistance mutations. We report our research efforts aimed at discovering a novel chemotype of anti-HIV-1 agents with improved potency against a variety of NNRTI-resistance mutations in this paper. Structural modifications of the lead <b>K-5a2</b> led to the identification of a potent inhibitor <b>16c</b>. <b>16c</b> yielded highly potent anti-HIV-1 activities and improved resistance profiles compared with the approved drug etravirine. The co-crystal structure revealed the key role of the water networks surrounding the NNIBP for binding and for resilience against resistance mutations, while suggesting further extension of <b>16c</b> toward the NNRTI-adjacent site as a lead development strategy. Furthermore, <b>16c</b> demonstrated favorable pharmacokinetic and safety properties, suggesting the potential of <b>16c</b> as a promising anti-HIV-1 drug candidate.

Currently, there has been considerable interest in the discovery of original molecules with broad-spectrum anti-HIV activity and favourable pharmacokinetic profiles, to be used as an alternative to the approved anti-HIV/AIDS drugs, should they fail as therapeutics. Five-membered azole heterocycles represent an important class of lead structures for novel anti-HIV drug development. They can serve as versatile building blocks to introduce different new functional groups, (i) as scaffolds to anchor these groups into the optimal space for interactions with the target, (ii) as basic pharmacophore elements to make hydrogen bonds or hydrophobic interaction for facilitating the spatial filling at the binding site, (iii) as ester surrogates to improve metabolic stability, or (iv) as pharmacophoric motif of metal coordination to coordinate metal ions (i.e. magnesium) within the active site of target (i.e. integrase). This article will summarize recent progress in the development of some azoles derivatives that inhibit the replication of HIV-1 and will illustrate the possible functional role(s) of the azole motif in the search for new anti-HIV drugs.
