This paper shows that proteins display an unexpectedly wide range of behaviors in buffers containing moderate (0.1-10 mM) concentrations of SDS (complete unfolding, formation of stable intermediate states, specific association with SDS, and various kinetic phenomena); capillary electrophoresis provides a convenient method of examining these behaviors. Examination of the dynamics of the response of proteins to SDS offers a way to differentiate and characterize proteins. Based on a survey of 18 different proteins, we demonstrate that proteins differ in the concentrations of SDS at which they denature, in the rates of unfolding in SDS, and in the profiles of the denaturation pathways. We also demonstrate that these differences can be exploited in the analysis of mixtures.
Fault intersections are a geometric complexity that frequently occurs in nature. Here we focus on earthquake rupture behavior when a continuous planar main fault has a second fault branching off of it.We use the finite element (FE) method to examine which faults are activated and how the surrounding material responds for both elastic and elastic-plastic off-fault descriptions.Compared to an elastic model, a noncohesive elastic-plastic material, intended to account for zones of damaged rock bordering maturely slipped faults, will inhibit rupture on compressional side branches and promote rupture of extensional side branches.Activation of extensional side branches can be delayed and is triggered by continued rupture propagation on the main fault. We examine the deformation near the branching junction and find that fault opening is common for elastic materials, especially for compressional side branches. An elastic-plastic material is more realistic because elevated stresses around the propagating rupture tip and at the branching junction should bring the surrounding material to failure. With an elastic-plastic material model, fault opening is inhibited for a range of realistic material parameters. For large cohesive strengths, opening can occur, but with material softening, a real feature of plastically deforming rocks, opening can be prevented. We also discuss algorithmic artifacts that may arise due to the presence of such a triple junction. When opening does not occur, the behavior at the triple junction is simplified and standard contact routines in FE programs are able to properly represent the physical situation.
We analyze the growth of cavities along grain interfaces by the combined processes of grain boundary diffusion and plastic dislocation creep in the adjoining grains. It is shown that the coupling between the processes can be expressed in terms of a parameter L, which has the dimensions of length and which is a function of material properties, temperature and applied stress; L decreases with increasing temperature and stress and has, e.g., values in the range of 0.25 to 25 μm for various pure metals when stressed to 10−3 × shear modulus at 0.5 Tm. The contribution of dislocation creep to the cavity growth rate is shown to be negligible when L is comparable to or larger than the cavity spacing, but significant interactions occur, leading to growth rates very much in excess of those predicted on the basis of boundary diffusion alone, when L is comparable to or smaller than the cavity size. The coupling occurs because extensive dislocation creep allows local accommodation of matter diffused into the grain boundary from the cavity walls. The cavity growth rate is analyzed by formulating a new variational principle that governs combined processes of grain boundary diffusion and non-linear viscous creep, and by implementing this principle through the finite-element method to obtain numerical solutions. Results for the cavity growth rate are presented for a wide range of ratios of L to cavity spacing, and of cavity radius to spacing. Also, results are presented for the total growth time of cavities from an initial size to final coalescence.
By way of numerical examples, this paper explores the nature of solutions to a class of strain gradient plasticity theories that employ conventional stresses, equilibrium equations and boundary conditions. Strain gradients come into play in these modified conventional theories only to alter the tangent moduli governing increments of stress and strain. It is shown that the modification is far from benign from a mathematical standpoint, changing the qualitative character of solutions and leading to a new type of localization that is at odds with what is expected from a strain gradient theory. The findings raise questions about the physical acceptability of this class of strain gradient theories.
