Reduced energy expenditure for a given energy intake level causes positive energy balance and eventually may lead to excess body weight and obesity. Two kinds of genetic effects are considered in this chapter. The first one is additive genetic effect, or the so-called heritability, and the second one is the genotype-environment interaction effect. Resting Metabolic Rate is a complex phenotype associated with the metabolic rates of all tissues and organs of the body measured in the basal state after an overnight fast. Although the mechanisms linking high fat consumption to increased body fat stores remain to be elucidated, inter-individual differences in substrate oxidation, particularly lipid oxidation, are likely to be involved. The thermic effect of food is the integrated increase of energy expenditure after food ingestion. Negative or positive energy balance sustained for a long period influences energy expenditure. The studies reviewed here suggest that inter-individual differences observed in various energy expenditure components are partly determined by the genotype.

The concern about the current epidemic of obesity is heightened by recent data showing an increase in the prevalence of overweight and obesity in children and adolescents. As obese children are at increased risk of becoming obese adults, early identification of individuals at risk of becoming obese is important for the implementation of preventive measures. In that context, it is important to assess the role of genetic and nongenetic factors in childhood obesity as well as in susceptibility to dietary variations. This paper reviews recent data regarding the prevalence of obesity in childhood as well as the role of genetic factors in phenotypes related to obesity, with emphasis on studies conducted on children, when available. The existence of severe cases of obesity, such as those encountered in Mendelian syndromes of obesity or those caused by mutations in single genes, provides strong evidence for a role of genetic factors in the aetiology of childhood obesity. Furthermore, results from family studies suggest that the maximal heritability of obesity phenotypes ranges from about 30% to 50% and that the major affectors of body fat content, energy intake and energy expenditure are also influenced by genetic factors. Finally, there is increasing evidence that responsiveness to dietary intervention is genetically determined.
Despite some evidence that genotype-environment interaction (G x E) effects may be involved in the variation observed in behavioral and biological traits, few attempts have been made to detect and quantify this component of genetic variation in humans. We propose that one way to achieve this goal is to challenge several genotypes in a similar manner, submitting both members of several MZ twin pairs to an ethically acceptable experimental treatment capable of inducing an adaptative response. In this situation, the G x E effect can be assessed with a two-way analysis of variance for repeated measures on one factor, the treatment effect. In this design, twins are considered nested within the pair, whereas the treatment effect is considered a fixed variable. The intrapair resemblance in the response to the treatment is quantified with an intraclass correlation coefficient computed with between-sibhips and within-sibhips means of squares. To illustrate this approach, changes induced by long-term endurance training were studied in 10 MZ twin pairs. Significant intrapair resemblance in the response of maximal oxygen uptake was observed, with about 7 to 8 times more variance between pairs than within pairs. This design with MZ twins may be helpful in the study of human variation for multifactorial phenotypes.

As a first step, based on problem constraints, we identified and retained three water treatment systems alternatives. In close collaboration with the stakeholders, we then defined and obtained weights for criteria that take into account public health, costs, system impacts and perception issues. The ELECTRE II method was used to aggregate the alternatives evaluations in order to obtain a ranking of the three water treatment systems. The process revealed that the system which combines NF and UV disinfection is ranked first for all the different stakeholder weightings. The sensitivity analysis of the aggregation parameters and evaluations reinforced that conclusion. SummaryFamily resemblance in fatness has been studied in 481 individuals from 114 families of French descent living in the greater Quebec city area. Subjects were measured for the following fatness indicators: triceps, biceps, subscapular, suprailiac, abdominal and medial calf skinfolds. The sum of these six skinfolds as well as a prediction of percent body fat (Durnin and Rahaman 1967) were also considered. Data were standardized for appropriate age and sex classes yielding SS scores. The influence of relevant lifestyle variables (energy intake, energy expenditure and socioeconomic status) were statistically removed from SS yielding residual scores (RS) that were then submitted to familial analyses. Analyses of variance indicate that there is a larger between family variation than within (P ⩽ 0·01) for SS when considering either the whole nuclear family or sibships; in these instances, the intra-class correlation ranges from 0·15 to 0·26. There was, however, no significant resemblance among spouses for the SS fatness indicators. Similar values were essentially found for RS fatness indicators. Furthermore, husband-wife inter-class correlations were not significant with the exception of subscapular and calf skinfold RS. Covariations between biological relatives are however significant (0·16 ⩽ r ⩽ 0·24, P ⩽ 0·01) for SS and remain essentially unchanged after statistical control over the lifestyle variables (0·16 ⩽ r ⩽ 0·40, P ⩽ 0·01). The findings that spouses do not covary significantly in fatness, while biological relatives of traditional nuclear families exhibit a significant degree of resemblance even after statistical control over daily energy intake, daily energy expenditure and socioeconomic status provide evidence supporting the hypothesis that there is a substantial genetic effect in human fatness.ZusammenfassungDie Familienähnlichkeit der Fettheit wurde untersucht bei 481 Individuen von 114 Familien französischen Ursprungs in der Großregion von Quebec. Die Summe dieser sechs Hautmaße wurde ebenso wie eine Schätzung des prozentualen Körperfetts (Durnin und Rahaman 1967) berücksichtigt. Die Daten wurden auf die entsprechenden Altersund Geschlechtsklassen standardisiert, mit dem Ergebnis von SS-Ziffern. Der Einfluß bedeutsamer Variablen des Lebensstils (Energieaufnahme, Energieverbrauch und sozioökonomischer Status) wurde statistische aus SS entfernt, womit Restziffern (RS) verblieben, die dann der Familienanalyse unterworfen wurden. Varianzanalysen zeigen für SS eine größere Zwischen- als Binnenfamilien varianz (P ⩽ 0,01), wenn man entweder die ganze Kernfamilie oder die Geschwister berücksichtigt; in diesen Fällen variiert die Intraklass-Korrelation von 0,15 bis 0,26. Les sujets ont été mesurés pour les indicateurs d'adiposité suivants: plis cutanés tricipital, bicipital, sous-scapulaire, supra-iliaque, abdominal et médian du mollet. La somme de ces 6 plis cutanés ainsi qu'une prédiction du pourcentage de graisse corporelle (Durnin et Rahaman 1967) ont aussi été considérées. Les données ont été standardisées pour des classes d'âge et de sexe appropriées, produisant des cotes de SS. L'influence de variables impliquées du style de vie (consommation d'énergie, dépense d'énergie et statut socio-économique) a été extraite statistiquement des SS, produisant des cotes résiduelles (RS) qui ont alors été soumises à des analyses familiales. Les analyses de variance indiquent qu'il y a une variation plus grande entre familles que dans les familles (P ⩽ 0,01) pour les SS que soient considérées la famille nucléaire entière ou les fratries; dans ces cas, la corrélation intra-classe s'étend de 0,15 à 0,26. Il n'y avait pas, cependant, de ressemblance significative entre époux pour les indicateurs SS d'adiposité. Des valeurs essentiellement semblables ont été trouvées pour les indicateurs RS d'adiposité. De plus, les corrélations mari-femme interclasses n'étaient pas significatives à l'exception des RS des plis cutanés sous-scapulaire et du mollet. Les covariations entre apparentés biologiques sont cependant significatives (0,16 ⩽ r ⩽ 0,24; P ⩽ 0,01) pour les SS et restent essentiellement inchangées après contrôle statistique des variables de style de vie (0,16 ⩽ r ⩽ 0,40; P ⩽ 0,01). Les résultats que les époux ne covarient pas significativement en adiposité, tandis que les apparentés biologiques de familles nucléaires traditionnelles montrent un degré significatif de ressemblance même après contrôle statistique de la consommation journalière d'énergie, de la dépense journalière d'énergie et du statut socio-économique apportent un support à l'hypothèse qu'il y a un effet génétique substantiel dans l'adiposité humaine.
