Activation-induced cytidine deaminase (AID) is required for immunoglobulin (Ig) gene class switch recombination (CSR), somatic hypermutation (SHM), and somatic hyperconversion. In general, high AID expression is found in mature B cells that are responding to antigens. However, AID expression and SHM have also been detected in developing B cells from transgenic mice that have a limited Ig repertoire. Here we demonstrate that AID expression, ongoing CSR, and active SHM occur in developing B cells from wild-type mice. Further, our results suggest that somatic variants arising from developing B cells in the bone marrow further diversify in the spleen of unimmunized mice. AID expression in developing B cells is T cell independent but involves engagement of B cell receptors and Toll-like receptors. Early AID expression can increase the preimmune repertoire of developing B cells, may provide an innate population of IgG- and IgA-expressing cells, and could be involved in receptor editing of self-reactive immature B cells.
ウイルスや細菌のもつ核酸(DNAとRNA)は自然免疫系により認識され,I型インターフェロンや炎症性サイトカインが産生され,感染病原体に対する生体防御応答が誘導される.我々は発現スクリーニングにより,二重鎖DNAに対する自然免疫応答を制御する細胞内分子としてTRIM56を同定した.TRIM56はユビキチンリガーゼとして機能し,STINGと呼ばれるアダプター分子のK63型ユビキチン化を促進した.この修飾により,TBK1キナーゼがリクルートされ最終的にI型インターフェロンが誘導された.以上のことから,DNAに対する自然免疫応答において,TRIM56によるユビキチン化を軸とした新たなシグナル伝達経路の存在が明らかとなった.一方,Toll-like receptor (TLR) 7とTLR9は,ウイルスの核酸を認識し,プラズマ細胞様樹状細胞からI型インターフェロン産生をさせる.我々は,抗ウイルス因子として報告されていたViperinが,プラズマ細胞様樹状細胞においてTLR7/9を介したI型インターフェロン産生に重要な役割を果たしていることを見出した.Viperinは,プラズマ細胞様樹状細胞においてTLR7/9の刺激より転写因子Interferon regulatory factor (IRF) 7依存的に強く誘導され,脂肪滴に局在している.Viperinは,プラズマ細胞様樹状細胞においてTLR7/9の下流で働きIRF7を活性化するシグナル伝達因子として知られているTRAF6とIRAK1に結合し,これらの因子を脂肪滴上へとリクルートする.その結果,IRAK1のK63結合型ユビキチン化が効率的に誘導され,IRAK1によるIRF7の活性化を介したI型インターフェロンの産生が促進される.Viperinが,直接的なウイルス複製阻害に加えて,TLR7/9を介したI型インターフェロン産生の促進により抗ウイルス応答に関わっていることが判明した.
The kinase TAK is required for the development of conventional and regulatory T cells. We previously reported that mice with conditional deletion of TAK1 in T cells (Lck-cre:TAK1fl/fl mice) exhibited severe T lymphopenia, and were nevertheless predisposed to spontaneous colitis with unknown etiology. Here we focused on the immunopathological mechanism in colitic Lck-cre:TAK1fl/fl mice. We found that ‘leaky’ CD4+ T cells retaining TAK1 acquired inflammatory phenotypes that contribute to disease onset in Lck-cre:TAK1fl/fl mice. Furthermore, the gut microbiota-triggered signaling was also a key event leading to the pathogenesis. We discovered that Lck-cre:TAK1fl/fl mice were almost completely devoid of TCRαβ+CD8α+ intestinal intraepithelial lymphocytes (IELs) and this was largely due to the developmental defect of the thymic precursors by TAK1 deficiency. Remarkably, transfer of TCRαβ+CD8α+ IELs from wild-type mice ameliorated colitis in Lck-cre:TAK1fl/fl mice. Taken together, our current study highlighted the emerging role of TAK1 in configuring the gut-specialized T cell subset, which regulates mucosal homeostasis under lymphopenic conditions.
