There is now a much better understanding the signalling pathways and cellular events involved in ageing, including evidence of cellular senescence with telomere shortening, activation of phosphoinositide 3-kinase(PI3K)-mammalian target of rapamycin (mTOR) signalling, impaired autophagy, mitochondrial dysfunction, stem cell exhaustion, epigenetic changes and abnormal microRNA profiles1.

Abstract The past 15 years have seen the introduction or major revision of structural stainless steel design codes throughout the world, and at the same time, interest in the use of stainless steel in construction has been accelerating. Historically the high initial material cost of stainless steel has limited its use primarily to specialist and prestige applications. However, the emergence of design codes, a better awareness of the additional benefits of stainless steel and a transition towards sustainability are bringing more widespread use into conventional structures. Although a number of similarities between stainless steel and ordinary carbon steel exist, there is sufficient diversity in their physical properties to require separate treatment in structural design. In addition to the straightforward differences in basic material properties (such as Young's modulus and yield strength), further fundamental differences exist, such as the nature of the stress–strain curve and the material's response to cold‐work and elevated temperatures; these have implications at ultimate, serviceability and fire limit states. This paper describes the use of stainless steel as a structural material, discusses current structural design provisions, reviews recent research activities and highlights the important findings and developments.
The effect of a potent inhibitor of angiotensin‐converting enzyme, ramipril, was studied on both inhaled histamine and bradykinin‐induced bronchoconstriction in six male, normotensive, mild asthmatic subjects. Oral administration of 10 mg ramipril caused no change in lung function or airway reactivity to inhaled histamine or bradykinin despite achieving adequate reduction in angiotensin‐converting enzyme activity.
The Xeltis biorestorative transcatheter heart valve (BTHV) leaflets are made from an electrospun bioabsorbable supramolecular polycarbonate-urethane and are mounted on a self-expanding nitinol frame. The acute haemodynamic performance of this BTHV was favourable.We sought to demonstrate the preclinical feasibility of a novel BTHV by evaluating the haemodynamic performances of five pilot valve designs up to 12 months in a chronic ovine model.Five design iterations (A, B, B', C, and D) of the BTHV were transapically implanted in 46 sheep; chronic data were available in 39 animals. Assessments were performed at implantation, 3, 6, and 12 months including quantitative aortography, echocardiography, and histology.At 12 months, greater than or equal to moderate AR on echocardiography was seen in 0%, 100%, 33.3%, 100%, and 0% in the iterations A, B, B', C, and D, respectively. Furthermore, transprosthetic mean gradients on echocardiography were 10.0±2.8 mmHg, 19.0±1.0 mmHg, 8.0±1.7 mmHg, 26.8±2.4 mmHg, and 11.2±4.1 mmHg, and effective orifice area was 0.7±0.3 cm2, 1.1±0.3 cm2, 1.5±1.0 cm2, 1.5±0.6 cm2, and 1.0±0.4 cm2 in the iterations A, B, B', C, and D, respectively. On pathological evaluation, the iteration D demonstrated generally intact leaflets and advanced tissue coverage, while different degrees of structural deterioration were observed in the other design iterations.Several leaflet material iterations were compared for the potential to demonstrate endogenous tissue restoration in an aortic valve in vivo. The most promising iteration showed intact leaflets and acceptable haemodynamic performance at 12 months, illustrating the potential of the BTHV.
